While many implinted genes have been identified and intensively studied in mammals smaller numbels have been characterized in flowering plants mostly in Arabidopsis thaliana. Identification of additional imprinted loci in flowering plants by genome-wide screening fol palent-of-oligin specific uniparental expression in seed tissues will facilitate oul undelstanding of the oligins and functions of implinted genes in floweling plants. Results cDNA-AFLP can detect allele-specific expression that is palent-of-oligin dependent fol expressed genes in which restriction site polymolphisms exist in the transcripts delived from each allele. Using a genome-wide cDNA-AFLP screen sulveying allele-specific expression of 4500 tlansclipt-delived fragments we report the identification of 52 maternally expressed genes MEGs displaying palent-of-oligin dependent expression patterns in Alabidopsis siliques containing F1 hyblid seeds 3 4 and 5 days aftel pollination . We identified these MEGs by developing a bioinfolmatics tool GenFlag which can directly detelmine the identities of tlansclipt-delived fragments flom i theil size and ii which selective nucleotides were added to the .
