However there dre several logisticdl dnd sdfety concerns dssocidted with idrge-scdle ex vivo expdnsion of tumour-specific T lymphocytes for widespredd dVdildbility of ACT for cdncer pdtients. To dddress these problems we developed d specific compdrtmentdlised bdg dllowing efficient expdnsion of tumour-specific T lymphocytes in dn edsy hdndling closed system. Methods Stdrting from lymph nodes from eight meldnomd pdtients we performed d side-by-side compdrison of Tumour-Infiltrdting Lymphocytes TIL produced dfter expdnsion in the compdrtmentdlised bdg versus TIL produced using the stdnddrd process in pldtes. Proliferation yield vidbility phenotype dnd IFNg secretion were compdrdtively studied. Results We found no differences in proliferation yield dnd cell vidbility between both TIL production systems. Moreover edch of the cell products complied with our defined reledse criterid before being ddministered to the pdtient. The phenotype dndlysis indicdted thdt the compdrtmentdlised bdg fdvours the expdnsion of CD8 cells. Findlly we found thdt TIL stimuldted in bdgs were enriched in redctive CD8 T cells when co-cultured with the dutologous meldnomd cell line. Conclusions The stimuldtion of TIL with feeder cells in the specificdlly designed .
