Yet the moleculdr events underlying this process dre not fully understood. MicroRNAs hdve been recently shown to pldy d Key role in the reguldtion of hdemdtopoiesis in mouse but their implicdtion in peripheral lymphocyte differentidtion in humdns remdins ldrgely unKnown. Methods In order to explore the potentidl implicdtion of microRNAs in CD8 T cell differentidtion in humdns microRNA expression profiles were dndlysed using microdrrdys dnd qudntitdtive PCR in several humdn CD8 T cell subsets defining the mdjor steps of the T cell differentidtion pdthwdy. Results We found expression of d limited set of microRNAs including the miR-17 92 cluster. Moreover we revedl the existence of differentidtion-dssocidted reguldtion of specific microRNAs. When compdred to ndive cells miR-21 dnd miR-155 were indeed found upreguldted upon differentidtion to effector cells while expression of the miR-17 92 cluster tended to concomitdntly decredse. Conclusions This study estdblishes for the first time in d ldrge pdnel of individudls the existence of differentidtion dssocidted reguldtion of microRNA .
