In vitro micropropagation from immature embryos of the endemic and endangered Muscari muscarimi Medik

An efficient in vitro bulblet production procedure from immature zygotic embryos of endemic and endangered Muscari muscarimi Medik. was described in the current study. Zygotic embryos were first isolated from immature seeds and cultured on different nutrient media compositions supplemented with various combinations of α-naphthalene acetic acid (NAA), picloram, dicamba, 6-benzylaminopurine (BAP), and thidiazuron (TDZ). | Turkish Journal of Biology Turk J Biol (2014) 38: 83-88 © TÜBİTAK doi: Research Article In vitro micropropagation from immature embryos of the endemic and endangered Muscari muscarimi Medik. 1, 2 3 4 5 Satı UZUN *, İskender PARMAKSIZ , Serkan URANBEY , Semra MİRİCİ , Ercüment Osman SARIHAN , 3 6 7 7 7 Arif İPEK , Mehmet Demir KAYA , Bilal GÜRBÜZ , Neşet ARSLAN , Cengiz SANCAK , 7 7 Khalid Mahmood KHAWAR , Sebahattin ÖZCAN 1 Department of Field Crops, Seyrani Agricultural Faculty, Erciyes University, Kayseri, Turkey 2 Department of Molecular Biology and Genetics, Faculty of Science and Arts, Gaziosmanpaşa University, Tokat, Turkey 3 Department of Biology, Faculty of Science, Çankırı Karatekin University, Çankırı, Turkey 4 Department of Biology Education, Gazi Education Faculty, Gazi University, Ankara, Turkey 5 Department of Field Crops, Faculty of Agriculture and Natural Sciences, Uşak University, Uşak, Turkey 6 Department of Field Crops, Faculty of Agriculture, Eskişehir Osmangazi University, Eskişehir, Turkey 7 Department of Field Crops, Faculty of Agriculture, Ankara University, Ankara, Turkey Received: Accepted: Published Online: Printed: Abstract: An efficient in vitro bulblet production procedure from immature zygotic embryos of endemic and endangered Muscari muscarimi Medik. was described in the current study. The best bulblet regeneration (59 bulblets per explant) was achieved in Murashige and Skoog (MS) medium containing 4 mg/L BAP and mg/L NAA after 1 year of culture initiation. Regenerated bulblets were then transferred into MS medium without plant growth regulators for rooting. Bulblets
