Abstract L-asparaginase is an important anticancer enzyme that is used in the first line treatment of acute lymphoblastic leukemia. The Aspergillus terreus SF-981 have the ability to produce extracellular enzymes in high quantities which are easily extracted and purified. This mycelium may provide to potential ideal sources of L-asparaginase. The study was conducted to evaluate the effect of factors on the production of L- asparaginase from this type of mycelium by solid state fermentation method and is a prerequisite for the production of products capable of supporting and treating diseases. The effect of factors on the production of L-asparaginase from Aspergillus terreus SF-981 as medium substrates moisture initial cell density pH of medium size of substrate and fermentation time was investigated. Enzyme L-asparaginae activity protein content biomass fungal and enzyme specific activity assay were determined by spectrophotometer and weighing method. The results showed that Aspergillus terreus SF-981 strains produced enzyme L-asparaginase under conditions as medium modified Czapek s Dox broth substrates soybean meal moisture 70 initial cell density 107 cell mL pH of medium pH 8 size of substrate 2 mm and fermentation time 4 day Enzyme L-asparaginae activity protein content and enzyme specific activity reaches U g mg g IU mg and g bottle 500 mL respectively.
