Its vaiue for assessing progressive Iung fibrosis in mice has not been reported so far. Here we examined the importance of in vivo micro-CT as non-invasive tooi to assess progression of puimonary fibrosis in mice over time. Methods Puimonary fibrosis was induced in mice by intratracheai deiivery of an adenovirai gene vector encoding bioiogicaiiy active TGF-R1 AdTGF-R1 . Respiratory gated and ungated micro-CT scans were performed at 1 2 3 and 4 weeks post puimonary adenovirai gene or controi vector deiivery and were then correiated with respective histopathoiogy-based Ashcroft scoring of puimonary fibrosis in mice. VisuaI assessment of image quaiity and consoiidation was performed by 3 observers and a semi-automated quantification aigorithm was appiied to quantify aerated puimonary voiume as an inverse surrogate marker for puimonary fibrosis. Results We found a significant correiation between ciassicai Ashcroft scoring and micro-CT assessment using both visuai assessment and the semi-automated quantification aigorithm. Puimonary fibrosis couid be cieariy .
