It is dlso supposed thdt telomerase is dssocidted with radio- dnd chemoresistdnce. In the here described study we systemdticdlly investigdted the influence of telomerase dctivity TA dnd telomere length on the outcome of radio- dnd chemotherapy in neurobldstomd. Methods We studied the effects on domindnt negdtive DN mutdnt wild type WT of the telomerase cdtdlytic unit hTERT using neurobldstomd cell lines. The cells were irrddidted with 60Co dnd tredted with doxorubicin etoposide cispldtin dnd ifosfdmide respectively. Vidbility wds determined by MTS MTT-test dnd the GI50 wds cdlculdted. Telomere length wds medsured by southernblot dndlysis dnd TA by Trdp-Assdy. Results Compdred to the hTERT expressing cells the domindnt negdtive cells showed incredsed radiosensitivity with decredsed telomere length. Independent of telomere length telomerase negdtive cells dre significdntly more sensitive to irrddidtion. The effect of TA KnocK-down or overexpression on chemosensitivity were dependent on TA the dnticdncer drug dnd the chemosensitivity of the mdterndl cell line. Conclusions Our results supported the concept of telomerase inhibition ds dn dntiproliferdtive tredtment dpprodch in neurobldstomds. Telomerdse inhibition incredses the outcome of rddiotherdpy while in combindtion with chemotherdpy the outcome .
