The lacrimal fluid samples were absorbed directly from the eyes with absorbing paper trips and then the absorbed DF was extracted therefrom by a mixture of phosphate buffer 0 01 M pH 2 5 and methanol 15 85 v v . Finally the obtained extracts were analysed by reverse-phase HPLC in observation to USP 41 NF 36 Monographs NPG 2018 with HPLC system - Agilent 1200 Series Agilent Technologies - Germ. Column - C18 x 250 mm Packed particle size - 5 μm Temperature - 35 oC . The method was validated by the US-FDA and ICH guidance and proposed for bioanalytical evaluation. The method proved evidently potent for quantification of DF in the rabbit lacrimal fluid at 6 minutes after administion of a commercial DF eye drop products of the kind. The developed method showed viable as a useful approach for the quality control of DF eye drop products and further development of new DF ophthalmic dosage forms.
